Zmiany : 

• W 2016 sezonie zobaczymy powrót Austrian Grand Prix, wielki powrót do MotoGP po 19 latach nieobecności tego toru, będzie się działo ! Zawody odbędą się na Red Bull Ring. 
• Tor w Indianapolis wypadł z MotoGP, był w zawodach od 2008r.
• Pierwszy raz zdarzy się że Duth TT odbywać się będzie w niedziele.

Zmiany Teamowe: 

• Marc VDS Racing dodają 2 motocykl.
• Po dodaniu 2 motocykli w 2015, Team LCR wystawia jedna maszynę do startów w 2016 r.
• Aspar Racing Team kończy współpracę z Honda i powraca do marki Ducati których motocykli używali w latach 2010-11.
• AB Motoracing odchodzi z MotoGP.
• Ioda Racing również odchodzi z  MotoGP, rozpoczynają starty w  World Superbike.
